Cette avanc\u00e9e pourrait ouvrir la voie \u00e0 un \u00e9mail fabriqu\u00e9 en laboratoire qui pourrait \u00eatre utilis\u00e9 dans les r\u00e9parations dentaires et pourrait m\u00eame permettre des obturations vivantes ou des dents vivantes compl\u00e8tement nouvelles pouvant \u00eatre implant\u00e9es dans la m\u00e2choire d&rsquo;un patient.  Les chercheurs ont \u00e9tudi\u00e9 l\u2019activit\u00e9 g\u00e9n\u00e9tique qui se produit au cours du d\u00e9veloppement dentaire, puis ont utilis\u00e9 ces informations pour inciter les cellules souches \u00e0 devenir des am\u00e9loblastes, qui sont le type de cellule responsable de la cr\u00e9ation de l\u2019\u00e9mail.  Une fois pr\u00e9sentes dans les organo\u00efdes, les cellules peuvent produire trois prot\u00e9ines essentielles \u00e0 l\u2019\u00e9mail.<\/p>\n<p>R\u00e9parer les dents est difficile.  Dans le pass\u00e9, les gens les faisaient simplement retirer et utilisaient \u00e0 la place des fausses dents.  Heureusement, la dentisterie moderne est un peu plus sophistiqu\u00e9e, mais implique rarement la r\u00e9g\u00e9n\u00e9ration des tissus dentaires endommag\u00e9s ou malades, et implique g\u00e9n\u00e9ralement le retrait et le remplacement des tissus par des alternatives synth\u00e9tiques.  M\u00eame si cela est acceptable, ce serait bien de pouvoir remplacer les dents manquantes par une alternative vivante ou de boucher un trou dans une dent avec du v\u00e9ritable \u00e9mail dentaire, plut\u00f4t qu&rsquo;avec une p\u00e2te polym\u00e8re synth\u00e9tique.    <\/p>\n<p>Cr\u00e9er des traitements r\u00e9g\u00e9n\u00e9ratifs pour le travail dentaire est un objectif noble, mais il se heurte \u00e0 un d\u00e9fi majeur, du moins dans le cas de l&rsquo;\u00e9mail des dents.  Lors de la formation des dents, l\u2019\u00e9mail est cr\u00e9\u00e9 par des cellules appel\u00e9es am\u00e9loblastes.  Cependant, ces cellules ne restent pr\u00e9sentes que pendant la formation des dents et meurent ensuite.  Par cons\u00e9quent, les dents n\u2019ont aucun moyen de r\u00e9g\u00e9n\u00e9rer leur propre \u00e9mail et il n\u2019existe pas de population endog\u00e8ne de telles cellules \u00e0 cibler chez les patients adultes.  Les chercheurs \u00e0 l\u2019origine de cette derni\u00e8re \u00e9tude ont r\u00e9alis\u00e9 que s\u2019ils souhaitaient cr\u00e9er de l\u2019\u00e9mail dentaire naturellement, de telles cellules devraient alors \u00eatre cr\u00e9\u00e9es \u00e0 partir de z\u00e9ro.  Et c&rsquo;est ce qu&rsquo;ils ont fait.<!-- ASYNC UAT M - 300x250(t,r) - --><\/p>\n<p><!-- END ASYNC UAT M - 300x250(t,r) --><\/p>\n<p>Les chercheurs ont utilis\u00e9 un outil appel\u00e9 s\u00e9quen\u00e7age d\u2019ARN \u00e0 indexation combinatoire unicellulaire (sci-RNA-seq) pour \u00e9tudier quels g\u00e8nes sont activ\u00e9s et d\u00e9sactiv\u00e9s \u00e0 chaque \u00e9tape du d\u00e9veloppement dentaire, leur donnant ainsi un plan de d\u00e9veloppement des am\u00e9loblastes.  Ensuite, ils ont activ\u00e9 ces m\u00eames g\u00e8nes, dans le bon ordre, pour amener les cellules souches indiff\u00e9renci\u00e9es \u00e0 se d\u00e9velopper en am\u00e9loblastes, qu\u2019ils ont cultiv\u00e9s sous forme d\u2019organo\u00efdes.  \u00c9tonnamment, les organo\u00efdes ont produit des prot\u00e9ines essentielles au d\u00e9veloppement de l\u2019\u00e9mail, indiquant une premi\u00e8re \u00e9tape sur la voie des traitements dentaires r\u00e9g\u00e9n\u00e9ratifs.  <\/p>\n<p>\u00ab De nombreux organes que nous aimerions pouvoir remplacer, comme le pancr\u00e9as, les reins et le cerveau humains, sont volumineux et complexes.  Les r\u00e9g\u00e9n\u00e9rer en toute s\u00e9curit\u00e9 \u00e0 partir de cellules souches prendra du temps \u00bb, a d\u00e9clar\u00e9 Hannele Ruohola-Baker, chercheuse impliqu\u00e9e dans l\u2019\u00e9tude.  \u00ab Les dents, en revanche, sont beaucoup plus petites et moins complexes.  Ce sont peut-\u00eatre les fruits les plus faciles \u00e0 trouver.
