La m\u00e9thode permet aux chercheurs de cr\u00e9er des images 3D haute r\u00e9solution de tissus c\u00e9r\u00e9braux vivants qui r\u00e9v\u00e8lent sa complexit\u00e9 cellulaire, et m\u00eame son \u00e9volution dans le temps, permettant de surveiller la plasticit\u00e9 neuronale.<\/p>\n<div class=\"wp-block-image\">\n<figure class=\"alignright size-full\"><img decoding=\"async\" loading=\"lazy\" width=\"283\" height=\"345\" data-lazy-type=\"image\" src=\"https:\/\/www.linkidoc.fr\/blog\/wp-content\/uploads\/2023\/08\/1691446071_184_Une-technique-dimagerie-revele-le-tissu-cerebral-vivant-dans-sa.jpg\" alt=\"\" class=\"lazy lazy-hidden wp-image-1550790\"  data-lazy-\/><noscript><img decoding=\"async\" loading=\"lazy\" width=\"283\" height=\"345\" src=\"https:\/\/www.linkidoc.fr\/blog\/wp-content\/uploads\/2023\/08\/1691446071_184_Une-technique-dimagerie-revele-le-tissu-cerebral-vivant-dans-sa.jpg\" alt=\"\" class=\"wp-image-1550790\"  \/><\/noscript><\/figure>\n<\/div>\n<p>La technique repose sur des approches d&rsquo;apprentissage en profondeur qui aident \u00e0 affiner la qualit\u00e9 de l&rsquo;image et \u00e9galement \u00e0 distinguer les structures cellulaires au sein de ce tissu dense et tr\u00e8s complexe.  L&rsquo;approche peut fournir des d\u00e9tails sans pr\u00e9c\u00e9dent sur les tissus vivants sans causer de dommages, car elle collecte le moins d&rsquo;informations possible, puis utilise l&rsquo;apprentissage en profondeur pour compl\u00e9ter les informations manquantes.  Sans cette \u00e9tape, la quantit\u00e9 de lumi\u00e8re n\u00e9cessaire pour obtenir une image aussi d\u00e9taill\u00e9e ferait frire le tissu vivant.<\/p>\n<p>Le cerveau est un organe extr\u00eamement complexe, contenant environ 86 milliards de neurones.  Ces cellules sont dispos\u00e9es en r\u00e9seaux sophistiqu\u00e9s et leur interconnectivit\u00e9 et leur activit\u00e9 sont \u00e9galement plastiques, \u00e9voluant dans le temps.  \u00c0 ce jour, il a \u00e9t\u00e9 tr\u00e8s difficile de capturer cette complexit\u00e9 dans des images haute r\u00e9solution, en particulier sans endommager ou autrement compromettre le tissu, ce qui rend impossible la cr\u00e9ation de telles images de tissus vivants au fil du temps.<!-- ASYNC UAT M - 300x250(t,r) - --><\/p>\n<p><!-- END ASYNC UAT M - 300x250(t,r) --><\/p>\n<p>Les approches pr\u00e9c\u00e9dentes utilis\u00e9es pour imager les tissus c\u00e9r\u00e9braux incluent la microscopie \u00e9lectronique, qui peut capturer des images haute r\u00e9solution, mais n\u00e9cessite que les tissus soient physiquement sectionn\u00e9s pour l&rsquo;imagerie 3D, ce qui est en quelque sorte un non-non pour les cerveaux vivants.  Les techniques de microscopie optique peuvent fournir une imagerie tridimensionnelle, mais g\u00e9n\u00e9ralement avec une r\u00e9solution m\u00e9diocre.<\/p>\n<p>Une approche plus r\u00e9cente est appel\u00e9e Super-resolution Shadow Imaging, qui, en combinaison avec une m\u00e9thode appel\u00e9e Stimulated Emission Depletion, peut fournir des images haute r\u00e9solution qui commencent \u00e0 transmettre la complexit\u00e9 cellulaire du cerveau, mais elle ne convient pas aux tissus vivants, car le quantit\u00e9 de lumi\u00e8re n\u00e9cessaire endommagerait encore les tissus vivants.<\/p>\n<figure class=\"wp-block-image size-full\"><img decoding=\"async\" loading=\"lazy\" width=\"770\" height=\"256\" data-lazy-type=\"image\" src=\"https:\/\/www.linkidoc.fr\/blog\/wp-content\/uploads\/2023\/08\/1691446072_888_Une-technique-dimagerie-revele-le-tissu-cerebral-vivant-dans-sa.jpg\" alt=\"\" class=\"lazy lazy-hidden wp-image-1550791\"  data-lazy-\/><noscript><img decoding=\"async\" loading=\"lazy\" width=\"770\" height=\"256\" src=\"https:\/\/www.linkidoc.fr\/blog\/wp-content\/uploads\/2023\/08\/1691446072_888_Une-technique-dimagerie-revele-le-tissu-cerebral-vivant-dans-sa.jpg\" alt=\"\" class=\"wp-image-1550791\"  \/><\/noscript><\/figure>\n<p>Cette nouvelle approche est con\u00e7ue pour fournir des images 3D haute r\u00e9solution qui transmettent la complexit\u00e9 cellulaire sans cuire les tissus.  Il utilise juste assez de lumi\u00e8re pour capturer les informations minimales requises pour qu&rsquo;un algorithme d&rsquo;apprentissage en profondeur fournisse les d\u00e9tails manquants.  Les r\u00e9sultats sont spectaculaires.  <\/p>\n<p>\u00ab Avec LIONESS, pour la premi\u00e8re fois, il est possible d&rsquo;obtenir une reconstruction compl\u00e8te et dense du tissu c\u00e9r\u00e9bral vivant.
